He earned a Bachelor of Laws from Cairo University in 1967, followed by a Postgraduate Diploma in Public Law (1969) and another in Administrative Sciences (1970), both from Cairo University. In January 1970, he was appointed as an Assistant Delegate to the State Council.

In 1973, he was seconded as a legal advisor to the General Authority for the Ministry of Foreign Affairs Buildings Financing Fund. He later served as an advisor to the General Secretariat (Legislative Secretariat) of the Council of Ministers in 1982. He was promoted to Assistant Advisor of Category (A) in 1980, then to Advisor to the State Council in 1984, Undersecretary of the State Council in 1990, and Vice President of the State Council in 1992.

On May 19, 2013, the General Assembly of the Supreme Constitutional Court appointed Counselor Adly Mansour as its President, succeeding Counselor Maher El-Beheiry. On July 3, 2013, he was appointed interim President of Egypt as President of the Supreme Constitutional Court, and took the constitutional oath on July 4, 2013. He led the country during the transitional period from July 4, 2013, to June 8, 2014.

Following his term, President Abdel Fattah El-Sisi honored him with the Grand Nile Necklace, the highest state honor, in the first presidential decree of his inauguration ceremony.
